Design Notes

Endogenously, this promoter controls the expression of Factor Induced Gene 1, which is transcribed upon activation by mating factor. For our project, this promoter was used to create an inducible effector feedback loop in the MAPK mating pathway. Upon activation and transduction of the MAPK pathway, any genes under the control of the Fig1 promoter will be transcribed. By putting a negative effector under this control, mating induced transcription will lead to decreased pathway output once the effector has been translated.

Source

endogenous promoter for Fig1 (Factor Induced Gene 1, Gene ID: YBR040W). Integral membrane protein required for efficient mating; may participate in or regulate the low affinity Ca2+ influx system, which affects intracellular signaling and cell-cell fusion during mating
